In response to the killing of the leader of the Islamic Republic, the General Staff of the Armed Forces of Iran threatened the U.S. and Israel, stating that they would "make them regret with power and firmness" and continue the path of the slain leader of Iran.
The Revolutionary Guards also stated in another announcement that they would "stand firm against internal and external conspiracies and punish the aggressors against the Islamic homeland."

State media quoted Mohammad Mokhber, assistant to Ali Khamenei, reporting that following the killing of the leader of the Islamic Republic, the responsibility for the transitional period will fall on the President, the Head of the Judiciary, and one of the jurists of the Guardian Council.
According to these reports, Mokhber indicated that the three senior officials would be responsible for managing the transitional period after the killing of the leader of the Islamic Republic.

ستاد کل نیروهای مسلح ایران در واکنش به کشته‌شدن رهبر جمهوری اسلامی، آمریکا و اسرائیل را تهدید کرد که «باقدرت، صلابت پشیمان» خواهد کرد و راه رهبر کشته‌شده ایران را ادامه خواهد داد.
سپاه پاسداران هم در بیانیه دیگری اعلام کرد که «در مقابله با توطئه های داخلی و خارجی و مجازات درس آموز متجاوزان به میهن اسلامی ایستادگی خواهند کرد.»

رسانه‌های حکومتی به نقل از محمد مخبر، دستیار علی خامنه‌ای، گزارش دادند که پس از کشته شدن رهبر جمهوری اسلامی، مسئولیت دوره انتقال بر عهده رئیس‌جمهور، رئیس قوه قضاییه و یکی از فقهای شورای نگهبان خواهد بود.
بر اساس این گزارش‌ها، مخبر با اشاره به سازوکار پیش‌بینی‌شده برای شرایط فوق‌العاده، اعلام کرده است که این سه مقام ارشد وظیفه مدیریت دوره انتقالی پس از کشته شدن رهبر جمهوری اسلامی را بر عهده خواهند داشت.
